Ganciclovir sodium salt equivalent to 500mg
ganciclovir in 10ml vial: i.v injection.
Mode of action: Ganciclovir is an antiviral drug.
It works by blocking the action of a viral
component called DNA polymerase. This
compound ·is required by the virus for it to copy
genetic material ( from RNA to DNA), a process
that is needed for the virus to multiply & survive.
Ganciclovir is also incorporated into the viral
DNA. By blocking the action of DNA
polymerase & disrupting virul DNA, ganciclovir
prevents CMV from multiplying.
Ind: 1. Treatment of cytomegalovirus (CMV)
infections in immunocompromised individuals.
2. Prevention of cytomegalovirus (CMV) disease,
in patients receiving immunosuppressive therapy
secondary to organ transplantation.
A/R: In patients ·who were being treated with
ganciclovir the most common haematological
side effects were neutropenia, anaemia and
thrombocytopenia.

Precautions: Ganciclovir i.v injection should not
be administered if the absolute neutrophil count
is less than 500 cells/ml. Because of the mutagenic
and teratogenic potential of ganciclovir,
ANTIMICROBIAL DRUGS
women of childbearing potential should be
advised to use effective contraceptive during
treatment. Similarly, men should be advised to
practice barrier contraceptive during and for at
least 90 days following treatment with
ganciclovir.
Dosage & admin: Treatment of CMV infection:
Initial (induction) treatment: 5mg/kg
infused at a constant rate over 1 hour every 12
hours (lOmg/kg/day) for 14 to 21 days. Longterm
(maintenance) treatment: For immunocompromised
patients at risk of relapse of
CMV retinitis- a course of maintenance
therapy may be given. I.V infusion of 6mg/kg
once daily, 5 days per week, or 5mg/kg once
daily, 7 days per week is recommended.
Prevention of CMV disease: Induction
regimen: Smg/kg infused every 12 hours.

(10mg/kg/day) for 7 to 14 days. Maintenance
regimen: l.V infusion of 6mg/kg once daily, 5
days per week, or Smg/kg once daily, 7 days
per week is recommended.
